I have a HTML5 game that is set to autoplay on page load, https://incal.itch.io/shuffle, are interactions with the iframe included in "Browser Plays" within Analytics, maybe via some clever mechanism? Or does it only include mobile users who had to click to load the game?

I don’t see any reason why not. You also get analytics on how many people went to your game page, so using that is a safe assumption.
