Execllent questions!

1.  As long as you are describing exactly what is going into the room and where, and what is being taken away, these types of changes are fine, as long as you have room in your quest notes - the clarity of this explanation and how many steps it takes may affect accessibility rating.

2.  Iconography is limited to the game system and chosen expansion, and letters/numbers (native icons in HQuestBuilder); to avoid bending the rule about only one expansion, I would use one of these unused letter/numbers or place a quest notes by the affected door.

3.  If the assets needed can be covered by the First Light box, then First Light should be the game system version chosen.

Before finalizing the jam page, I forgot to take inventory and see if FL was actually missing anything present in the Game System.  I actually need to simplify this language on the jam page.

I believe at least as far as the board assets are concerned in the AH version of the GS and FL there is parity, ie the same number and type of assets other than the dragon, the underground board and the furniture as tiles (which could potentially be used in new ways).  I did an inventory of the game components that I think is accurate (though only the design components- not the cards so there will be differences there as well iirc).  I didn't inventory my MB game as I'm pretty sure I have some extra tiles in there so I couldn't rely on it.  The big difference there should just be the blank black on the double walls, and I think the stair tile unless they changed the count on the pairings for the single tiles.  Getting a better count on the max number of walls or other features when combining with a single expansion was a major reason I inventoried them anyway.
