They can't do a trial run because that 1 dose is all there is. The other kids are all dead, Oz doesn't have the samples or ability to get more. It's just that 1, which is why it's a 'use it or don't' choice. They can't use the vault to see if it works or not because the vault relies on trauma, and dying from sickness while sad wouldn't be traumatic enough for anyone.
