It depends on the engine you're using. You don't normally chop the image up.  Most engines support tilesets so you can select portions of the tileset as "tiles" to use.

Don't save it as any compressed format, such as JPEG, that is where you lose image quality (due to compression). Most used file formats to retain quality in most engines are *.PNG files.
