Can some please explain what is the difference and benefits of having more “Virility”, “Focus” and “HP” and how to use them best in a DUAL? Also, the Gray padlocks that are shown on some challenged villagers pages, is there more to them?

Virility is in regards to how much "damage" you do in duels - so the higher virility the more effective your actions will be, focus is how fast your little head moves across the bar, so higher focus and the head will move slower making it easier to get a perfect hit. HP is just HP, you can last longer in duels and the enemy will require more actions to beat you. 

As for your last question, I think its in regards to affinity? The last locked square doesn't open till end game, where you get to choose the villager you want to be with.
