what do i do after the yusuke quest where i give him the key? he gave me a map but i dont know how to get to the lab

oh man. i gotta restart the whole game cause i dont have a save :(

how do i do the deed with anne? i did the collecting ingredients quest and saw the npcs talking about meeting anne, but i've tried going behind the tavern at every possible time over and over, but nothing ever happens. did i miss something?
