I have the same issue. Thanks for the tip on the ingredients. It's possible it's not a bug, but there's a specific time limit on the quest; if you don't complete it in time, it'll be destroyed. I'm not sure, it's just a guess.

It's probably impossible to free him, since we've already started her route. And during attempts to free her, she should be absent from the area, and when leaving the room, she only reminds us that the pregnancy is over, and nothing more can be done with words, she's stuck.

Yeah, we have no choice but to take Dr. Frost's path — the other doctor's route was never actually finished/implemented. You should be able to dig up Dr. Muramasa's reply in the main page threads/posts.
