Is it necessary to be alpha to enslave other inmate ,my reputation as alpha now is -1 and my slave is run away but i can't kidnap him back

You dont really need to he an alpha to kidnap someone, it just makes them easier to hold on the cell
If a slave runs away you gotta do the whole "imma slave you" process, yes even the little tasks, again
