Which one do you think is the better route and what content does both include?

Assuming you mean Helping Mario or Helping Sofia: 

Helping Sofia has more story content (and possibly more future content with Kat and Emma) and is one of the trophies in the game. Betraying Sofia to Mario allows you to keep fucking Kat & Emma without finishing their content and also unlocks the final room for sex scenes with a mindbroken Sofia.

So for now it's up to you whether you want more shared scenes with Mario and a sex-crazed Sofia, or do you want the canon route and the trophy? (If you fuck Kat enough that she starts asking for sex on the terrace & convince Emma to NOT sleep with Mario, then you can still fuck them on either route)
